Rapid and Effective Isolation of Mouse Circulating Tumor Cells Using ScreenCell Technology for Preclinical Analysis


A recent study conducted at Paul Brousse Hospital in Villejuif, France, focused on the collection of blood samples from animal models for research purposes. The study used specific pathogen-free conditions and ethical guidelines to house and monitor the health of mice. Blood samples were collected from wild-type C57BL/6 mice for normal blood collection and PyMY transgenic mice with breast tumors. Tumor cell lines were prepared from 4T1 mouse breast cancer cells and MCF7 human breast cancer cells for further experimentation.

The study employed ScreenCell Cyto kits to isolate circulating tumor cells (CTCs) from blood samples. The kits were used to filter blood through a membrane and identify CTCs based on cytological criteria. The recovery rate of the ScreenCell technology was evaluated by spiking a known number of cells into blood samples and isolating them using the kit.

Genomic DNA was extracted from the captured cells for further analysis and quantification. The concentration of DNA was determined using a Qubit fluorometer. Additionally, CTCs from blood samples containing 4T1 cancer cells were cultured using the ScreenCell MB kit in DMEM medium for 10 days.

Overall, the study highlighted the use of animal models, blood collection techniques, and the isolation and culture of CTCs for research purposes. The ethical handling of animals and adherence to proper protocols were emphasized throughout the study, showcasing a commitment to ethical and responsible scientific practices.
